  • Correlation and prognostic value analysis of CCT5 and OMA1 expression with epithelial mesenchymal transition in gastric cancer tissue

    ObjectiveTo investigate the correlation between the expression of chaperonin containing TCP1 subunit 5(CCT5) and metalloproteinase associated protein 1(OMA1) in gastric cancer(GC) tissue and epithelial mesenchymal transition(EMT), and to analyze their clinical prognostic value.MethodsFrom February 2019 to February 2020,104 patients with GC diagnosed and treated in the Department of Gastroenterology of Wuhan Third Hospital were selected. The expression of CCT5 mRNA, OMA1 mRNA and EMT markers Twist mRNA, N-cad mRNA and Vimentin mRNA were detected by real-time fluorescence quantitative PCR; The expression of CCT5 and OMA1 protein was detected by immunohistochemistry; Pearson correlation analysis was used to analyze the correlation between CCT5 mRNA, OMA1 mRNA and EMT indexes; Kaplan-Meier curve and COX regression were used to analyze the effects of CCT5 mRNA and OMA1 mRNA on the prognosis of GC patients.ResultsCCT5 mRNA, OMA1 mRNA, Twist mRNA, N-cad mRNA and Vimentin mRNA in GC cancer tissues were higher than those in adjacent tissues(t/P=71.623/<0.001, 81.587/<0.001, 38.717/<0.001, 52.234/<0.001, 44.813/<0.001). The positive rates of CCT5 and OMA1 in GC cancer tissues were 86.54 %(90/104) and 88.46 %(92/104), which were higher than those in adjacent tissues 5.77 %(6/104) and 7.69 %(8/104)(χ2=136.500, 135.893,P<0.001). The expression of CCT5 mRNA and OMA1 mRNA in GC was positively correlated with Twist mRNA, N-cad mRNA and Vimentin mRNA(r/P=0.754/<0.001, 0.612/<0.001, 0.649/<0.001; 0.716/<0.001, 0.695/<0.001, 0.713/<0.001); The expression levels of CCT5 mRNA and OMA1 mRNA were increased in GC patients with worse differentiation, lymph node metastasis and higher TNM stage(CCT5:t/F/P=302.221/<0.001, 12.763/<0.001,244.850/<0.001;OMA1:t/F/P=402.771/<0.001,13.069/<0.001,305.421/<0.001); The 3-year overall survival rate of CCT5 mRNA high expression group was 44.00 %(22/50), which was lower than 75.93 %(41/54) of low expression group, the difference was statistically significant(Log-Rank χ2=7.156, P=0.008). The 3-year overall survival rate of OMA1 mRNA high expression group was 39.22 %(20/51), which was lower than 81.13%(43/53) of low expression group(Log-Rank χ2=8.901,P=0.003). Poor differentiation, lymph node metastasis, TNM stage Ⅲ, high CCT5 mRNA and high OMA1 mRNA were risk factors affecting the prognosis of GC patients[OR(95%CI)=1.554(1.217-1.986),1.670(1.274-2.189), 1.751(1.302-2.354), 1.495(1.105-2.021),1.483(1.079-2.037)].ConclusionThe expression of CCT5 and OMA1 in GC is up-regulated, which is related to EMT indicators and is a tumor marker for evaluating the prognosis of GC patients.
  • Study on the relationship between serum GPER1, SESN2 levels and diabetes retinopathy in patients with type 2 diabetes

    ObjectiveTo study the relationship between serum G protein coupled estrogen receptor 1(GPER1), sestrin 2(SESN2) levels and diabetic retinopathy(DR) in patients with type 2 diabetes mellitus(T2DM).MethodsA total of 162 patients with T2DM diagnosed and treated in the 940 Hospital of Joint Logistics Support Force from February 2020 to February 2022 were retrospectively selected as the T2DM group. According to the results of eye examination, T2DM patients were divided into DR subgroup(n=68) and non-DR subgroup(n=94). According to the severity of DR, DR patients were divided into proliferative subgroup(n=28) and non-proliferative subgroup(n=40). 60 cases of healthy physical examination in the same period were selected as the healthy control group. The levels of serum GPER1 and SESN2 were detected by enzyme-linked immunosorbent assay. Multivariate Logistic regression analysis was used to analyze the influencing factors of DR; the predictive value of serum GPER1 and SESN2 levels for DR was evaluated by receiver operating characteristic(ROC) curve.ResultsThe serum GPER1 level in the retinopathy group was higher than that in the healthy control group, and the serum SESN2 level was lower than that in the healthy control group(t/P=36.850/<0.001,28.112/<0.001); the incidence of DR in 162 patients was 41.96 %(68/162). The duration of diabetes, insulin resistance index(HOMA-IR) and serum GPER1 in DR subgroup were higher than those in non-DR subgroup, while glomerular filtration rate(eGFR) and serum SESN2 were lower than those in non-DR subgroup(t/P=11.552/<0.001, 3.078/0.009, 17.226/<0.001, 21.915/<0.001, 11.100/<0.001). The serum GPER1 level in the proliferative DR subgroup was higher than that in the non-proliferative DR subgroup, and the serum SESN2 level was lower than that in the non-proliferative DR subgroup(t/P=12.151/<0.001,3.534/<0.001); Multivariate Logistic regression results showed that long duration of diabetes, high HOMA-IR and high GPER1 were independent risk factors for DR[OR(95%CI)=1.237(1.096-1.397), 1.265(1.079-1.483), 1.347(1.054-1.721)], and high eGFR and SESN2 were independent protective factors for DR[OR(95%CI)=0.736(0.598-0.906),0.759(0.596-0.967)];The area under the curve(AUC) of serum GPER1, SESN2 levels and the combination of the two for predicting DR were 0.731, 0.710 and 0.820, respectively. The AUC of the combination of the two was greater than that of serum GPER1 and SESN2 levels alone(Z=4.711, 4.816,P<0.001).ConclusionSerum GPER1 increases and SESN2 decreases in DR patients. Both of them are involved in the occurrence and development of DR. The combination of the two has a high predictive value for the occurrence of DR.
  • Study on the relationship between serum VASH-1, HOTAIR expression and diabetes retinopathy

    ObjectiveTo investigate the relationship between serum Vasohibin-1(VASH-1) protein, homeobox transcript antisense RNA(HOTAIR) expression and diabetic retinopathy(DR).MethodsAccording to the ratio of 1∶1∶1, 60 patients with PDR(PDR subgroup), 60 patients with non-PDR(NPDR subgroup) and 60 patients with non-DR(NDR subgroup) admitted to the Department of Endocrinology in Shaanxi Provincial People 's Hospital from January 2021 to January 2023 were selected as the T2DM group, and 90 healthy subjects were selected as the healthy control group, the expression of serum VASH-1 and HOTAIR was detected by enzyme-linked immunosorbent assay and real-time fluorescence quantitative PCR; Spearman method was used to analyze the correlation between serum VASH-1, HOTAIR expression and blood glucose parameters in T2DM patients; Multivariate disordered Logistic regression was used to analyze the factors of DR in patients with T2DM;ROC curve was drawn to evaluate the predictive value of serum VASH-1 and HOTAIR expression for DR and PDR.ResultsCompared with the healthy control group, the expression of serum VASH-1 and HOTAIR in the T2DM group increased(t/P=14.646/<0.001, 14.203/<0.001). The expression of serum VASH-1 and HOTAIR in NDR subgroup, NPDR subgroup and PDR subgroup increased in turn(F/P=11.728/<0.001, 43.850/<0.001). The expression of serum VASH-1 and HOTAIR in T2DM patients was positively correlated with fasting blood glucose and glycosylated hemoglobin(HbA1c)(r/P=0.210/0.005, 0.458/<0.001, 0.220/0.003, 0.482/<0.001). The independent risk factors for the occurrence and development of DR in T2DM patients were long course of T2DM, high HbA1c, high VASH-1 and high HOTAIR[OR(95%CI)=3.108(2.110-4.579)/2.011(1.487-2.719), 6.634(2.761-15.940)/3.643(1.719-7.719), 1.996(1.427-2.792)/1.587(1.196-2.104), 2.600(1.683-4.015)/1.686(1.201-2.366)]. The AUC of DR and PDR predicted by the combination of the two was 0.869 and 0.886, respectively, which were higher than the AUC predicted by serum VASH-1 and HOTAIR expression alone, which were 0.803, 0.785, and 0.787, 0.778, respectively(Z/P=2.986/0.003, 3.230/0.001, 3.581/<0.001, 3.837/<0.001).ConclusionElevated serum VASH-1 and HOTAIR expression are independent risk factors for the occurrence and development of DR in T2DM patients. Serum VASH-1 combined with HOTAIR expression has a high value in predicting DR and PDR.
  • The expression levels of serum Endocan and ISM1 in patients with gestational diabetes mellitus and their relationship with adverse pregnancy outcomes

    ObjectiveTo study the expression of serum endothelial cell specific molecule(Endocan) and Isthmin1(ISM1) in patients with gestational diabetes mellitus(GDM) and their relationship with adverse pregnancy outcomes.MethodsA total of 94 patients with GDM diagnosed and treated in the Department of Obstetrics, the Fifth Affiliated Hospital of Xinjiang Medical University from January 2020 to January 2023 were selected as GDM group. According to the pregnancy outcome, they were divided into poor prognosis subgroup(36 cases) and good prognosis subgroup(58 cases), 48 healthy pregnant women who underwent physical examinations during the same period were selected as the healthy control group. The levels of serum Endocan and ISM1 were detected by enzyme-linked immunosorbent assay. Pearson correlation analysis was used to analyze the correlation between serum Endocan, ISM1 and clinical indicators. Logistic regression model and receiver operating characteristic curve were used to analyze the evaluation value of serum Endocan and ISM1 on adverse pregnancy outcomes in GDM patients.ResultsCompared with the healthy control group, the serum Endocan, fasting blood glucose(FPG), glycosylated hemoglobin(HbA1c), fasting insulin(FINS) and homeostasis model insulin resistance index(HOMA-IR) in the GDM group increased, and the serum ISM1 decreased(t/P=21.588/<0.001,12.762/<0.001,13.945/<0.001,12.337/<0.001,14.308/<0.001,20.599/<0.001); FPG, HbAlc, FINS and HOMA-IR were positively correlated with serum Endocan and negatively correlated with serum ISM1 in GDM group(r/P=0.632/<0.001,0.701/<0.001,0.698/<0.001,0.727/<0.001;-0.709/<0.001,-0.607/<0.001,-0.626/<0.001,-0.745/<0.001); Compared with the good prognosis subgroup, the poor prognosis subgroup had higher serum Endocan, HOMA-IR and lower serum ISM1(t/P=8.025/<0.001,7.988/<0.001,13.643/<0.001); High serum Endocan and HOMA-IR were risk factors for adverse pregnancy outcomes of GDM, and high ISM1 was a protective factor[OR(95%CI)=1.556(1.230-1.968),1.354(1.102-1.663),0.681(0.543-0.855)]; The AUC of Endocan, ISM1 and the combination of the two in predicting adverse pregnancy outcomes of GDM were 0.808, 0.821 and 0.890, respectively, and the combination of the two was better than the individual prediction efficiency(Z=4.515, 4.226, all P<0.001).ConclusionThe increase of serum Endocan and the decrease of serum ISM1 in GDM pregnant women are related to the body's glucose metabolism index. The combination of the two has a high evaluation value for the prognosis of pregnancy outcome in GDM patients.
  • The effect of Huahong Granules combined with Nifuratel Nystatin in the treatment of recurrent vulvovaginal candidiasis

    ObjectiveTo observe the therapeutic effect of Huahong Granules combined with nifuratelomycin vaginal soft gel in patients with recurrent mycotic vaginitis(VVC).MethodsFrom April 2022 to August 2023, 124 patients with relapsed VVC in our hospital were selected and divided by random lot method. 62 patients in the control group were given nifuratrol vaginal soft capsule treatment, and 62 patients in the experimental group were combined with Huahong granules based on the control group. The efficacy, TCM syndrome score, recurrence rate, vaginal microecology, inflammation level and adverse reactions were compared between the two groups.ResultsThe total effective rate of the experimental group was 88.71%, which was higher than the 74.19% of the control group(χ2/P=4.324/0.038). After treatment, the scores of itching in the genitals, excessive leucorrhea, and thick or beancurd-like discharge in the experimental group were lower than those in the control group, respectively(t/P=6.954/<0.001,5.906/<0.001,3.627/<0.001). After treatment, the colony density and flora diversity in the experimental group were better than those in the control group, and the vaginal pH value was lower than that in the control group(t/U/P=4.919/<0.001,3.041/0.002,2.978/0.003). After treatment, the levels of high-sensitivity C-reactive protein(hs-CRP) and granulocyte colony-stimulating factor(GM-CSF) in the experimental group were lower than those in the control group, respectively, while the level of transforming growth factor β1(TGF-β1) was higher than that in the control group(t/P=3.182/0.002,2.934/0.004,3.287/0.001). After 6 months of follow-up, the recurrence rate of the experimental group was 30.65%, which was lower than the 48.39% of the control group(χ2/P=4.083/0.043).There was no significant difference in the incidence of adverse reactions between the experimental group and the control group(P>0.05).ConclusionIn the treatment of recurrent VVC, Huahong Granules and nifuratel vaginal soft gel have significant effects, which can reduce recurrence, relieve clinical symptoms, improve vaginal microecology, reduce inflammation level, and have high safety.
