  • Expression and clinical significance of SPON2 and MGP in colorectal cancer tissue

    Objective To investigate the expression of spinal protein 2(SPON2) and matrix Gla protein(MGP) in colorectal cancer(CRC) tissue and their relationship with clinical pathological features and prognosis.Method Ninety-four CRC patients underwent surgery who were admitted to the General Surgery Department of Nanjing First Hospital from January 2018 to January 2020 were selected. Immunohistochemical staining was used to detect the expression of SPON2 and MGP in cancer and adjacent tissues; Analyze the relationship between the expression of SPON2 and MGP and the clinical pathological characteristics and prognosis of CRC patients; Multivariate Cox regression analysis of prognostic factors in CRC patients. Results The positive rates of SPON2 in CRC cancer and adjacent tissues were 65.96%(62/94) and 6.38%(6/94), respectively, while the positive rates of MGP were 63.83%(60/94) and 8.51%(8/94), respectively. The positive rates of SPON2 and MGP in CRC cancer tissues were higher than those in adjacent tissues(χ2=72.251, 62.298, both P<0.001). The positive rates of SPON2 and MGP in patients with TNM stage III, low differentiation, and lymph node metastasis were higher than those in patients with TNM stage I-II, high differentiation, and no lymph node metastasis, respectively(χ2/P=9.694/0.002, 10.883/0.001, 14.100/< 0.001, 14.785/< 0.001, 9.731/0.002, 12.739/< 0.001). The 3-year survival rates of SPON2 positive and negative groups were 54.84%(34/62) and 86.67%(26/30), respectively; The 3-year survival rates of MGP positive and negative groups were 53.33%(32/60) and 87.50%(28/32), respectively. The 3-year cumulative survival rate of SPON2 positive group and MGP positive group is lower than that of SPON2 negative group and MGP negative group(χ2=8.966, 12.420, P=0.003,<0.001). Tumor TNM stage III, low differentiation, lymph node metastasis, SPON2 positive, and MGP positive are independent risk factors affecting the prognosis of CRC patients [HR(95% CI)=1.768(1.226-2.551), 1.613(1.223-2.126), 1.600(1.167-2.194), 1.866(1.324-2.630), 1.315(1.151-1.503), all P<0.001].Conclusion Elevated expression of SPON2 and MGP in CRC is associated with adverse clinical and pathological features of CRC, and they are new tumor markers for evaluating the prognosis of CRC patients.
  • Early diagnostic value of serum IFN and YKL-40 levels for urinary tract infection after percutaneous nephrolithotomy

    Objective To explore the value of serum interferon-γ(IFN-γ) and chitinase protein-40(YKL-40) in the early diagnosis of urinary tract infection after percutaneous nephrolithotomy(PCNL). Methods From May 2018 to May 2023, 108 patients with PCNL who visited Beijing Jishuitan Hospital Guizhou Hospital were collected as the study subjects. They were separated into an infected group(n=41) and an uninfected group(n=67) based on whether they had urinary tract infection 24 hours after surgery. The clinical data of the two groups were compared; ELISA method was applied to detect and compare serum IFN and YKL-40 levels in two groups; Pearson method was applied to analyze the correlation between serum IFN and YKL-40 levels and indicators of urinary tract infection in patients with urinary tract infection after PCNL; Logistic regression was applied to analyze the influencing factors of urinary tract infections after PCNL; receiver operating characteristic(ROC) curve was applied to evaluate the diagnostic value of serum IFN and YKL-40 levels for urinary tract infections after PCNL. Results The operation time, the proportion of history of diabetes and the proportion of renal insufficiency in the infected group were obviously higher than those in the uninfected group [(t(χ2)/P=4.356/0.037, 5.016/0.025, 8.682/0.003)]; the levels of serum IFN and YKL-40 in the infected group were obviously higher than those in the uninfected group(t/P=7.276/<0.001, 7.161/<0.001); Pearson correlation analysis showed that there was a positive correlation between serum IFN and YKL-40 levels in patients with urinary tract infection after PCNL(P<0.05); Logistic regression analysis showed that serum IFN-γ levels were positively correlated with WBC and CRP, PCT, IL-6, and TNF-αlevels in patients with urinary tract infections complicated by PCNL(r/P=0.251/0.025, 0.428/0.004, 0.584/<0.001,0.632/<0.001, 0.374/0.008), and YKL-40 levels were positively correlated with WBC and CRP, PCT, IL-6, and TNF-α levels(r/P=0.603/<0.001, 0.459/0.002, 0.538/<0.001, 0.474/<0.001, 0.346/0.019); Logistic regression analysis showed that elevated levels of IFN-γand YKL-40 were independent risk factors affecting the complication of urinary tract infections after PCNL[OR(95%CI)=1.602(1.175-2.184), 1.665(1.207-2.296)]; ROC curve analysis results showed that the area under the curve(AUC) of serum IFN and YKL-40 levels alone and in combination for the diagnosis of PCNL complicated with urinary tract infections was 0.788, 0.782, and 0.877, respectively, the combined diagnostic effect of the two was better than that of single diagnosis(Z/P=2.114/0.035, 2.747/0.006). Conclusion The serum levels of IFN-γ and YKL-40 in patients with urinary tract infection after PCNL obviously increase, and the combined detection of the two has high diagnostic value for urinary tract infection after PCNL.
  • A study on the correlation between levels of fatty acid oxidation and lipolytic mediators and penile erectile dysfunction

    Objective To explore the correlation between levels of fatty acid oxidation and pro lipolytic mediators and penile erectile dysfunction. Methods Seventy patients with erectile dysfunction treated at the Urology Center of the First Affiliated Hospital of Xinjiang Medical University from October 2022 to April 2023 were selected as the observation group, and an additional 70 healthy males during the same period were selected as the healthy control group. Compare the general clinical data, serum testosterone(T), erectile hardness score(EHS), maximum systolic blood flow velocity(PSV) of unilateral penile artery, serum lipid peroxidation(LPO), expression levels of vasopressin D1(RvD1) and vasopressin E1(RvE1) between two groups of subjects. Screening for risk factors for erectile dysfunction through correlation analysis and multivariate logistic regression analysis. Further evaluate the predictive value of various risk factors for the occurrence of erectile dysfunction through receiver operating characteristic curve(ROC) and area under curve(AUC). Results The average serum LPO of the observation group patients was significantly higher than that of the healthy control group(t/P=8.714/<0.001), while RvD1, RvE1, EHS scores, and PSV levels were significantly lower than those of the healthy control group(t/P=10.109/<0.001, 10.410/<0.001, 6.271/<0.001, 13.243/<0.001); Pearson correlation analysis showed that serum LPO levels were negatively correlated with EHS and PSV(r/P=-0.275/0.033,-0.233/0.049), while RvD1 and RvE1 levels were positively correlated with EHS and PSV(r/P=0.391/<0.001, 0.366/<0.001, 0.423/<0.001, 0.387/<0.001); Multivariate logistic regression analysis showed that elevated serum LPO levels were independent risk factors for the occurrence of erectile dysfunction [OR(95%CI)=1.127(1.045-1.216)], and elevated levels of RvD1 and RvE1 were protective factors for the occurrence of erectile dysfunction [OR(95%CI)=0.761(0.669-0.866), 0.787(0.707-0.876)]; ROC curve analysis shows that serum LPO, RvD1, and RvE1 have high predictive power for the occurrence of erectile dysfunction, and their combined predictive value for disease is higher than the independent predictive value of each indicator(Z/P=1.873/0.013, 1.803/0.019, 1.711/0.033). Conclusion Elevated levels of serum fatty acid oxidation and decreased levels of pro lipolytic mediators are risk factors for the occurrence of erectile dysfunction, providing new ideas for further exploration of the mechanisms underlying the occurrence and development of erectile dysfunction.
  • The clinical value of serum TPO Ab, TG Ab, and TR Ab in predicting pregnancy outcomes in subclinical hypothyroidism with gestational hypertension

    Objective To analyze the expression levels of thyroid peroxidase antibody(TPO Ab), thyroglobulin antibody(TG Ab), and thyroid stimulating hormone receptor antibody(TR Ab) in the serum of patients with subclinical hypothyroidism(hypothyroidism) combined with gestational hypertension(preeclampsia), and their clinical predictive value for pregnancy outcomes. Methods Two hundred and forty-two pregnant women with gestational hypothyroidism who established fertility records, underwent prenatal examinations, and gave birth at the Northwest Women and Children's Hospital in Xi'an from August 2020 to March 2023 were selected. They were divided into 68 cases of gestational hypothyroidism group and 174 cases of hypothyroidism combined with gestational hypertension group based on whether they were complicated with gestational hypertension. Additionally, 242 normal pregnant women of the same period were selected as the normal pregnancy group; Using electrochemiluminescence immunoassay to detect the expression levels of serum TPO Ab, TG Ab, and TR Ab; Multivariate logistic regression analysis of the influencing factors of adverse pregnancy outcomes in patients with gestational hypothyroidism combined with hypertension; The predictive value of receiver operating characteristic(ROC) curve analysis of serum TPO Ab, TG Ab, and TR Ab expression levels on pregnancy outcomes in patients with hypothyroidism combined with preeclampsia. Results The expression levels of serum TPO Ab, TG Ab, and TR Ab increased sequentially in the normal pregnancy group, gestational hypothyroidism group, and hypothyroidism combined with preeclampsia group(F/P=85.036/<0.001, 26.675/<0.001, 23.086/<0.001). The levels of thyroid stimulating hormone(TSH) in the normal pregnancy group, pregnant hypothyroidism group, and hypothyroidism combined with preeclampsia group increased sequentially(F/P=85.052/<0.001). Compared with patients with good pregnancy outcomes, patients with poor pregnancy outcomes showed significantly higher levels of systolic blood pressure, diastolic blood pressure, TSH, total cholesterol(TC), triglycerides(TG), low-density lipoprotein cholesterol(LDL-C), TPO Ab, TG Ab, and TR Ab expression, while high-density lipoprotein cholesterol(HDL-C) was significantly reduced, the differences were statistically significant(t/P=3.852/<0.001, 2.421/0.017, 2.244/0.026, 2.342/0.020, 2.835/0.005, 2.707/0.007, 3.475/0.001, 3.399/0.001, 2.636/0.009, 4.280/<0.001). Elevated systolic blood pressure, diastolic blood pressure, TPO Ab, TG Ab, TR Ab, TC, and LDL-C are risk factors for adverse pregnancy outcomes in patients with gestational hypothyroidism combined with hypertension [OR(95% CI)=3.819(1.376-10.603), 3.235(1.198-8.739), 5.917(2.341-14.953), 9.043(2.924-27.965), 1.768(1.252-2.496), 2.534(1.085-5.921), 4.010(1.559-10.314)]. The AUC of serum TPO Ab, TG Ab, and TR Ab, as well as their combined prediction of pregnancy outcomes in patients with gestational hypothyroidism combined with hypertension, were 0.723, 0.701, 0.735, and 0.837, respectively. The combined predictive efficacy of the three was better than their individual predictive efficacy(Z/P=2.317/0.021, 2.764/0.006, 2.107/0.035). Conclusion The expression levels of TPO Ab, TG Ab, and TR Ab in the serum of patients with hypothyroidism combined with preeclampsia were significantly increased. The combination of the three has higher value in predicting adverse pregnancy outcomes and can be used as a biological indicator for predicting pregnancy outcomes.
  • The effect of curcumin on the expression of IL-36α and IL-36γ in TNBS induced colitis in rats

    Objective To investigate the effect of curcumin on the expression of IL-36α and IL-36γ in colitis induced by trinitrobenzenesulfonic acid(TNBS) in rats.Methods The experiment was conducted in May 2022 at the Medical Experimental Animal Center of Guangxi Medical University. Forty male SD rats were randomly divided into a normal control group, a model group, a curcumin group, and a sulfasalazine group, with 10 rats in each group. The model group, curcumin group, and sulfasalazine group of rats were prepared with TNBS ethanol solution enema to create rat colitis models, while the normal control group was given an equal volume of 0.9% sodium chloride solution enema. The curcumin group and sulfasalazine group were orally administered with curcumin and sulfasalazine solutions of 100 mg·kg-1·d-1 from the second day of modeling. The normal control group and model group were orally administered with an equal volume of 0.9% sodium chloride solution daily; After 7 days of administration, the rats were euthanized and serum and colon mucosa were collected. The body weight, disease activity index(DAI), and intestinal mucosal injury index(CMDI) of each group of rats were compared; ELISA method was used to detect the serum levels of IL-36α and IL-36γ in rats, and immunohistochemical method was used to measure the protein expression of IL-36α and IL-36γ in intestinal mucosal tissue.Results Compared with the normal control group, the DAI and CMDI scores of the model group increased significantly(P<0.01); The levels of serum IL-36α and IL-37 both increased(P<0.01); Compared with the model group, the DAI, CMDI scores, and serum IL-36α and IL-36γ levels in both the curcumin group and the sulfasalazine group decreased, and the curcumin group was lower than the sulfasalazine group, with statistically significant differences(F=3.531, 3.842, 24.503, 36.435, P<0.001); Compared with the normal control group, the protein expression levels of IL-36 and IL-37 in the model group increased(P<0.05); Compared with the model group, the protein expression levels of IL-36α and IL-36γ in the colon mucosa of the curcumin group and the sulfasalazine group decreased, and the curcumin group was lower than the sulfasalazine group, with statistical significance(F=3.461, 3.524, P<0.001). Conclusion Curcumin can significantly inhibit the expression of IL-36α and IL-36γ in serum and colon mucosa of experimental colitis rats.
