Relationship between serum G6PD and IGF-1 levels with the severity of the disease and hearing loss in newborns with hyperbilirubinemia

ObjectiveTo explore and analyze the relationship between serum levels of glucose-6-phosphate dehydrogenase(G6PD) and insulin-like growth factor-1(IGF-1) with the severity of disease and hearing loss in neonates with hyperbilirubinemia.MethodsClinical data of 102 newborns with high bilirubin admitted to Department of Pediatric Medicine, Xinzhou District People's Hospital from December 2022 to December 2023 were collected and assigned into mild group(n=41), moderate group(n=33), and severe group(n=28) based on the severity of their condition. Serum G6PD and IGF-1 levels were detected using ELISA, the general clinical data of the children were collected, receiver operating characteristic(ROC) curve was plotted to analyze the diagnostic value of G6PD and IGF-1 in assessing the degree of hearing loss of the patient's condition, Logistic was applied to analyze the factors influencing the degree of hearing loss in children, Pearson method was applied to analyze the correlation between serum G6PD, IGF-1 levels, bilirubin level, and hearing thresholds in children.ResultsThe levels of G6PD and IGF-1 in the moderate and severe groups were obviously lower than those in the mild group(F/P=51.881/<0.001, 42.334/<0.001); the serum G6PD and IGF-1 levels of children in the moderate and severe subgroups were obviously lower than those in the mild subgroup(F/P=26.243/<0.001, 22.454/<0.001), G6PD and IGF-1 were negatively correlated with bilirubin level and hearing thresholds in children(G6PD:r/P=-0.421/<0.001,-0.405/<0.001, IGF-1:r/P=-0.437/<0.001,-0.422/<0.001); the AUC of serum G6PD, IGF-1, and their combination in the diagnosis of the degree of hearing loss of disease was 0.780, 0.800, and 0.884, respectively, the combined diagnosis was obviously better than the individual diagnosis of G6PD and IGF-1(Z=2.905, 2.109, P=0.004, 0.035); Bilirubin and UCB were independent risk factors for the degree of hearing impairment in children[OR(95% CI)=1.354(1.111-1.650), 1.157(1.022-1.309)], and G6PD and IGF-1 were independent protective factors for the degree of hearing impairment in children[OR(95%CI)=0.864(0.773-0.966), 0.864(0.773-0.966)].ConclusionIn newborns with hyperbilirubinemia, serum levels of G6PD and IGF-1 obviously decrease with the degree of hearing loss of disease, and both have auxiliary diagnostic value for the severity of disease, which are influencing factors for the degree of hearing loss in children.

Study on the correlation between serum HSP60, HSP70 and Th17/Treg cells and prognosis in children with primary immune thrombocytopenia

ObjectiveTo analyzed the relationship between serum heat shock protein 60(HSP60), heat shock protein 70(HSP70) and helper T cell 17/regulatory T cell(Th17/Treg) cells and prognosis in children with primary immune thrombocytopenia(ITP).MethodsOne hundred and twenty-eight ITP children diagnosed and treated in the Department of Pediatrics, Baoshan People's Hospital from June 2020 to June 2022 were selected as observation group, and 60 healthy children in the same period were selected as control group. The levels of serum HSP60, HSP70 and Th17/Treg cells were detected and compared between two groups. The correlation between serum HSP60, HSP70 and Th17/Treg cells was analyzed by Pearson method. ITP children were followed up for 1 year after discharge, and were divided into good prognosis subgroup and poor prognosis subgroup according to different prognosis. The clinical data of ITP children were collected, the influencing factors of prognosis in ITP children were analyzed by univariate and multivariate Logistic regression models, and the risk prediction nomogram model was constructed. The predictive value of serum HSP60, HSP70 and Th17/Treg cells on the prognosis of ITP children was analyzed by established receiver operating characteristic(ROC) curve.ResultsThe levels of serum HSP60, HSP70 and Th17/Treg cells in observation group were significantly higher than those in control group(t/Z/P=20.445/<0.001, 17.467/<0.001, 5.823/<0.001). Pearson analysis showed that, serum HSP60 and HSP70 were positively correlated with Th17/Treg cells(r/P=0.417/<0.001, 0.348/<0.001). multivariate="" logistic="" regression="" analysis="" showed="" high="" and="" treg="" levels="" were="" independent="" risk="" for="" poor="" prognosis="" in="" itp="" plt="" count="" at="" initial="" diagnosis="" was="" a="" protective="" .="" the="" results="" of="" roc="" curve="" auc="" level="" nomogram="" model="" 0.840="" 0.975="" respectively.="" bootstrap="" b="1" used="" to="" verify="" bias-corrected="" prediction="" basically="" coincident="" with="" ideal="" ability="" better.="" decision="" shows="" threshold="" probability="" range="" its="" net="" return="" rate="">0, which was higher than the two invalid lines.ConclusionThe levels of serum HSP60, HSP70 and Th17/Treg in IPT children are significantly increase, and serum HSP60 and HSP70 are positively correlated with Th17/Treg. PLT count, HSP60, HSP70 and Th17/Treg levels at initial diagnosis are the influencing factors of poor prognosis in ITP children, and the nomogram model based on independent risk factors such as HSP60 and HSP70 has a good predictive value for poor prognosis in ITP children.

The expression and prognostic value of serum LncRNA NEAT1 and TFF1 in retinoblastoma

ObjectiveTo investigate the expression and prognostic value of serum long chain non coding RNA nuclear enriched transcript 1(LncRNA NEAT1) and trefoil factor 1(TFF1) in children with retinoblastoma(Rb).MethodsEighty-five Rb patients who underwent ophthalmic surgery at Ophthalmology Department of Beijing Anzhen Hospital Affiliated to Capital Medical University Nanchong Hospital from January 2018 to January 2021 were selected as the Rb group, while 60 healthy individuals who underwent physical examinations during the same period were selected as the healthy control group. Real time fluorescence quantitative PCR was used to detect serum LncRNA NEAT1 levels, and enzyme-linked immunosorbent assay was used to detect serum TFF1 levels; the relationship between serum LncRNA NEAT1, TFF1 and clinical pathological characteristics was analyzed; the evaluation value of serum LncRNA NEAT1 and TFF1 in Rb patients was analyzed by job characteristic curve of subjects.ResultsThe serum LncRNA NEAT1 in the Rb group was(3.12 ± 0.43) higher than that in the healthy control group(0.82 ± 0.21), the serum TFF1 of the Rb group was(22.14 ± 4.61) μ g/L, which was lower than that of the healthy control group(64.01 ± 9.63) μ g/L(t/P=38.305/<0.001, 34.858/<0.001). Compared with patients with tumor diameter<20 mm and pathologically differentiated Rb, patients with tumor diameter ≥ 20 mm and undifferentiated Rb had higher serum LncRNA NEAT1 and lower TFF1(Lnc RNANEAT1:t/P=17.925/<0.001, 16.848/<0.001; TFF1:t/P=12.505/<0.001, 8.120/<0.001). The 3-year progression free survival rate of the high expression group of serum LncRNA NEAT1 was lower than that of the low expression group(57.50% vs. 88.89%), and the 3-year progression free survival rate of the low expression group of TFF1 was lower than that of the high expression group(57.14% vs. 90.70%)(Log rank χ2/P=13.551/<0.001, 15.310/<0.001); Pathologically undifferentiated type and high serum LncRNA NEAT1 were risk factors affecting the prognosis of Rb, while high serum TFF1 was a protective factor [HR(95%)CI=1.523(1.147-2.024), 1.473(1.108-1.957), 0.612(0.413-0.908)]; The area under the curve of serum LncRNA NEAT1, TFF1, and their combined prediction of Rb prognosis were 0.836, 0.861, and 0.921, respectively, which were larger than the single detection of serum LncRNA NEAT1 and TFF1(Z=4.823, 4.312,P<0.001).ConclusionSerum LncRNA NEAT1 levels increase and serum TFF1 levels decrease in Rb patients, both of which are related to the maximum tumor diameter and pathological classification, and are factors affecting the prognosis of Rb patients. The combination of the two can help evaluate the prognosis of patients.

Experimental study on the effects of epimedium sagittatum on bone metabolism and tibial bone microstructure in osteoporosis rats

ObjectiveTo investigate the effects of epimedium sagittatum(EPI) on bone metabolism and tibial bone microstructure in osteoporosis(OP) rats.MethodsRats were randomly separated into Sham group, Model group, EPI low-dose(EPI-L) group, EPI high-dose(EPI-H) group, and EPI-H+YC-1(HIF-1α inhibitor) group. Bilateral oophorectomy was performed to construct an OP rat model. ELISA was applied to detect serum IL-10, IL-6, TNF-α, serum calcium(Ca), serum phosphorus(P), osteoprotegerin(OPG), osteocalcin(OCN), and alkaline phosphatase(ALP) in rats. Dual energy X-ray bone density instrument was applied to detect tibial bone density(BMD) in rats. Micro CT was applied to detect the microstructure of tibia bone. HE staining was applied to observe the morphology of tibial tissue. Bone morphogenetic protein(BMP-2), RUNt-associated transcription factor 2(Runx2), and cysteine proteinase-3(cleaved) were detected by Western blot Protein levels of caspase-3, hypoxia inducible factor HIF-1α(HIF-1α), vascular endothelial growth factor(VEGF), and vascular endothelial growth factor receptor 2(VEGFR-2).ResultsCompared with the Sham group, the microstructure and tissue morphology of the tibia of rats in the Model group were obviously damaged, the ratio of tibial bone surface area to volume, trabecular separation, serum IL-6, TNF-α, ALP levels, and tibial bone tissue cleaved caspase-3 protein expression level were obviously increased(all P<0.05), the bone trabecular thickness, serum IL-10, OPG, OCN, S-Ca and S-P levels, tibial BMD, tibial bone tissue BMP-2, Runx2, HIF-1α, VEGF, and VEGFR-2 protein expression levels were obviously reduced(all P<0.05). Compared with the Model group, the damage to the microstructure and tissue morphology of the tibia of rats in the EPI-L and EPI-H groups was obviously reduced, the ratio of tibial bone surface area to volume, trabecular separation, serum IL-6, TNF-α, ALP levels, and tibial bone tissue cleaved caspase-3 protein expression level were obviously reduced(all P<0.05), the bone trabecular thickness, serum IL-10, OPG, OCN, S-Ca and S-P levels, tibial BMD, tibial bone tissue BMP-2, Runx2, HIF-1α, VEGF, and VEGFR-2 protein expression levels were obviously increased(all P<0.05). Compared with EPI-H group, the above indexes were partially reversed in EPI-H+YC-1 group(all P<0.05).ConclusionEPI can reduce inflammatory reaction, regulate bone metabolism balance, and improve tibial bone microstructure in OP rats, possibly by activating the HIF-1α/VEGF/VEGFR-2 signaling pathway.
